Research Description: 

The Wilcock lab is interested in vascular cognitive impairment and dementia; the second most common cause of dementia behind Alzheimer's disease. In addition to being a major cause of dementia, Alzheimer's disease patients commonly have vascular cognitive impairment as a co-morbidity. We have projects to examine the molecular mechanisms of vascular cognitive impairment, focusing primarily on inflammatory processes. We also have projects that determine the influence cerebrovascular disease has on the progression and severity of Alzheimer's disease, as well as how these vascular pathologies affects response to Alzheimer's disease targeted therapeutics. Finally, we have a project in collaboration with Elizabeth Head of Pharmacology and Frederick Schmitt of Neurology to assess neuroinflammatory changes in Down Syndrome. We are developing a translational research program examining neuroinflammatory proteins and homocysteine as modifiable biomarkers of cognitive impairment due to both Alzheimer's disease and cerebrovascular pathologies.

2R01 HD064993 Head/Schmitt (MPI) Aging and Dementia in Down syndrome: Connectivity, Inflammation and Cerebrovascular Contributions
The study will identify new ways in which to detect Alzheimer disease (AD) in adults with Down syndrome (DS) by monitoring changes in cognitive function, measuring brain changes by magnetic resonance imaging and profiling patterns of proteins in the plasma. In parallel, autopsy studies of brain samples will help us to understand how noninvasive cognitive, imaging and blood measures reflect the development of AD in DS.

1R01NS079637 - Vascular breakdown in Alzheimer’s Disease with cerebrovascular disease.
This is a grant to study the co-morbidity of vascular dementia and Alzheimer’s disease focusing on the neuroinflammatory mechanisms of both dementias and the therapeutic efficacy of targeting one dementia as opposed to the other.

DSDNIP-13-282631 - Inflammatory biomarkers to predict transition to dementia in Down Syndrome
A grant to study changes in inflammatory biomarkers in the plasma on Down Syndrome patients at risk of transitioning to dementia. We will look at markers pre and post transition to establish potential predictive biomarkers.
